Abstract
A fluorometric quantitative method for
determining flavonols was developed that uses boric acid to detect the chemical
structure R−CX−C−CO−C=C−R’ (X=O, OH, OCH3).
The total flavonol levels in the plasma, liver, and kidney of mice
administered orally with quercetin were determined by this analytical
method. The maximum excitation
wavelength of the fluorophore generated in the reaction with the flavonol, i.e.
quercetin, and boric acid was 460 nm and the maximum emission wavelength was
506 nm. The calibration curve for
quercetin was linear between 5.0 mg/mL and 0.50 mg/mL. Recoveries of quercetin from the
homogenates of mouse liver and kidney were good, at 80 ~ 90%, however, recovery
from the plasma was too high, at around 130%. In the plasma, liver and kidney of mice administered orally
with quercetin at doses of 100 and 200 mg/kg body weight, the concentration of
total flavonols 60 min after the administrations was dose dependent. These results suggest that this
analytical method could evaluate the concentration of total flavonols in the
plasma, liver, and kidney of mice administered orally with quercetin,
detecting the sum of the concentrations of the original compound and its
metabolites retaining the chemical structure R−CX−C−CO−C=C−R’ (X=O,
OH, OCH3).
